ABSTRACT
Background: Thyroid cancer (TC) is considered as the fastest growing malignancy in human endocrine system, particularly papillary thyroid (PTC). MicroRNAs (miRs) serve a role promoting or suppressing tumors various types of malignant tumor including PTC. This study aims to explore whether microRNA-26a-5p (miR-26a-5p) could affect proliferation, invasion and metastasis ability PTC cells by regulating Wnt5a. Materials methods: The expression miR-26a-5p was examined qRT-PCR tissue samples (58 cases, mean age 53 years old) cell lines (K1 BCPAP). Cell migration were tested with CCK8 assay, colony formation transwell assay wound healing respectively. Luciferase reporting experiment used verify that Wnt5a molecular target miR-26a-5p. relationship between analyzed Western blot further proved Pearson's correlation analysis. Animal (24 nude mice) experiments demonstrate inhibits growth targeting Results: declined tissues ( P <0.01). miR-26a-5 also significantly down-regulated advanced TNM stages <0.01) lymph node compared normal tissues. Compared line Nthy-ori 3-1, K1 BCPAP nearly 4.02-fold 2.51-fold reduced. Up regulation inhibited formation, cells. MiR-26a-5p negatively regulated r =−0.887, <0.01), yet overexpression reversed tumor-suppressive effect verified Conclusion: Overexpression depresses invasion, via Therefore, may represent potentially effective gene for Keywords: carcinoma, microRNA-26a-5p, Wnt5a,
