ABSTRACT
Introduction: Non-small cell lung cancer (NSCLC) is a worldwide malignance threatening human life. TGF-β/Smad signaling known to regulate proliferation, differentiation, migration and growth. As the only co-Smad playing crucial roles in TGF-β signaling, Smad4 reported be frequently mutated or occur as alternatively spliced tumor cells. was involved TGF-β-induced EMT process. However, whether alternative splicing occurs process NSCLC not clear. Methods: In our current study, we explored of during A549 10 ng/mL used induce EMT. Then, nest-PCR agarose electrophoresis were performed detect expression variants sequencing get variant DNA sequences. For recombinant cells, lentiviral infect order explore effects on proliferation MTT assay, colony formation assay wound-healing done. The E-cad VIM protein through Western blot. Results: There several novel gene fragments expressed results showed that they indeed reported. found have significant also regulated expression. Conclusion: Our indicated functional study these affect expression, showing potential targets for therapy. Keywords: Smad4, EMT, TGF-β, variants, non-small
