ABSTRACT
Purpose: This study aims to reveal the mechanism underlying baicalin-suppressing ovarian cancer stemness. Methods: OVCAR-3 and primary cells were used for cell model. The stem isolated by suspension culture. Cell viability clonogenicity examined CCK-8 assay colony formation assay. self-renewal of was evaluated determination sphere-forming capacity frequency in vitro vivo tumor-initiating cells. mRNA protein levels relatively quantified qRT-PCR Western blot. transcription regulation target genes tested luciferase reporter a modified nuclear rn-on Results: Treatment with non-toxic dose baicalin significantly inhibited spherogenicity Moreover, treatment suppressed Furthermore, expression markers (CD133 ALDH1A1) treatment. Baicalin inhibits YAP activity suppresses RASSF6, positive regulator YAP, at transcriptional level. Overexpression both RASSF6 abolished inhibitory effect on proliferation stemness Conclusion: results this demonstrated that attenuating via inhibiting finding revealed as novel inhibitor could serve an anti-cancer drug eradicating Keywords: baicalin, cancer, stemness,
