ABSTRACT
SRY-box containing gene 17 (SOX17) was reported to be a candidate tumor suppressor in multiple tumors. Little is known about its role clear-cell renal cell carcinoma (ccRCC). This study aims identify the epigenetic regulation and tumor-suppressive function of SOX17 ccRCC.Fifty-five human ccRCC tissue samples, ten adjacent non-malignant kidney 20 paired paraffin section tissues seven RCC lines were obtained. Immunohistochemistry (IHC) real-time PCR used examine expression target genes at mRNA protein levels. The methylation analyzed using methylation-specific (MSP) bisulfite genomic sequencing (BGS) assay. functions examined by CCK8, colony formation, wound healing assay Matrigel invasion assays. Luciferase analyze WNT signaling pathway. We investigated IHC. lower tissues. Next, we DNA promoter 55 tissues, 10 MSP. region occurred 60% 10% In vitro experiments showed that suppressed proliferation cells. Furthermore, inhibited migration cells as shown addition, found overexpression affected pathway downregulating c-myc cyclinD1.In summary, our downregulated loss regulated via mechanisms ccRCC. negatively regulates
