ABSTRACT
Aim: DNA hypermethylation is an epigenetic mechanism which induces silencing of tumor-suppressor genes in prostate cancer. Many studies have reported that specific components food plants like soy phytoestrogens may protective effects against carcinogenesis or progression. Genistein and daidzein, the major phytoestrogens, been to ability reverse cancer cell lines. The aim this study was investigate potential demethylating these two compounds on BRCA1, GSTP1, EPHB2 BRCA2 promoter genes. Methods & materials: Prostate lines DU-145 PC-3 were treated with genistein 40 µM, daidzein 110 budesonide (methylating agent) 2 µM 5-azacytidine (demethylating µM. In human we performed methylation quantification by using Methyl Profiler analysis. This technique based a methylation-specific digestion followed quantitative PCR. We analyzed corresponding protein expression western blotting.
