Chondrogenic differentiation of induced pluripotent stem cells from osteoarthritic chondrocytes in alginate matrix

ABSTRACT
Induced pluripotent stem cells (iPSCs) have the potential to revolutionise cell therapy; however, it remains unclear whether iPSCs can be generated from human osteoarthritic chondrocytes (OCs) and subsequently induced differentiate into chondrocytes.In present study, we investigated differentiation of OCs using defined transcription factors explored possibility these OC-derived for chondrogenesis.Our study demonstrates that are indistinguishable embryonic (hESCs).To promote chondrogenic differentiation, used lentivirus transduce seeded in alginate matrix with transforming growth factor-β1 (TGF-β1) then vitro co-cultured chondrocytes.Gene expression analysis showed this combinational strategy promotes established matrix.Increased cartilage-related genes, including collagen II, aggrecan, cartilage oligomeric protein (COMP), decreased gene degenerative marker, vascular endothelial factor (VEGF), were observed.The histological results revealed a dense sulphated extracellular co-culture TGF-β1-transfected matrix.Additionally, vivo chondroinductive activity was also evaluated.Histological examination more new formed matrix.Taken together, our data indicate by defi ned significantly improved chondrogenesis iPSCs.This work adds understanding solutions replacement problem.
