ABSTRACT
To enhance the keratinolytic potential of bacterial strain Bacillus cereus N14, isolated from poultry farm Sundernagar, District Mandi, Himachal Pradesh, different process parameters like temperature, pH, incubation time, inoculum size, age etc. were optimized using One Variable at a Time (OVAT) approach and Response Surface Methodology (RSM). N14 showed maximum enzyme activity (19.5 U/mL) after 3rd day 35°C, pH 9.0, 12.5 (%) size 3 days old culture 2.5 2.0 per cent maltose yeast extract as best carbon nitrogen source, respectively in presence Mn2+ divalent metal ion EDTA media additive. Central composite design (CCD) RSM was successfully applied to investigate effect 5 independent variables viz., time (days), inoculums (%), concentration source on keratinase production. The (74.86 attained by B. 22nd run. main highlight present study is that significant increase 27.5 production optimizing parameters, observed.
