ABSTRACT
A possible role of transporter associated with antigen processing (TAP)-l in the pathogenesis IDDM has been investigated by examining level TAP-1 expression islets pancreas and studying vitro effect interferon (IFN)-γ, IFN-α, tumor necrosis factor-α cultured islet cells. remarkable hyperexpression found endocrine cells (β non-β) islets, which constitutes first evidence this molecule target organ an autoimmune disease. correlated clearly HLA class I but only very partially II ectopic expression. IFN-γ both cytokines putatively implicated pathogenesis, were capable inducing protein (as assessed immunofluorescence flow cytometry) message (by Northern blot analysis reverse transcription polymerase chain reaction). These findings suggest that under influence (most probably IFN-α) β-cells may express their surface a high density I–peptide complexes facilitate recognition lysis low-affinity CD8+ T-cells.
