ABSTRACT
Abstract Introduction: GAD2 gene encodes the glutamate decarboxylase enzyme which catalyses transformation of into γ-aminobutyric acid, GABA. It is suggested that some polymorphic alleles gene, such as -243A>G, have an increased transcriptional effect compared with wild type, results in increase GABA hypothalamus subsequent neuropeptide Y, thus exacerbating hunger centre and appetite. The aim this study was to observe association between -243A>G polymorphism obesity, comparatively studying a group obese patients normal weight. Patients method: 127 were clinically evaluated Genetic Endocrine Department Children’s Emergency Clinical Hospital, Cluj. included two groups, case group, obesity (BMI higher than 97 kg/m2) control weight less kg/m2). Genotyping for GAD2-243A>G performed using PCR-RFLP technique, groups being regarding genotypes phenotypes. Results conclusions : In there statistically significant difference BMI (kg/m2) subgroups different (p=0.01), AA genotype severely affected AG GG genotypes. no (AA, or GG). Also, greater distribution G allele odds ratio suggest risk factor development (GG OR=3.76, OR=1.73, p=0.04). finding our important explaining multifactorial model research demonstrating GAD2-243 A> variant could be added other obesogenic factors would potentiate their effect.
