MiRNA-27a-3p promotes osteogenic differentiation of human mesenchymal stem cells through targeting ATF3.

ABSTRACT
To elucidate whether miRNA-27a-3p can promote osteogenic differentiation of hMSCs by targeting ATF3, thus alleviating osteoporosis symptoms.The serum levels in patients (n=20) and normal controls were detected quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). Human bone marrow mesenchymal stem cells (hMSCs) subjected to for 1, 3 7 days. Subsequently, mRNA miRNA-27a-3p, ALP, Bglap determined qRT-PCR. The regulatory effects the Bglap, Runx2 detected. After overexpression or knockdown we evaluated changes alizarin red staining ALP staining. Through Dual-Luciferase Reporter Gene Assay, verified binding relationship between ATF3. Rescue experiments finally conducted prove regulated ATF3.The level remained lower relative controls. With prolongation differentiation, gradually increased. upregulated protein osteogenesis-related genes, increased activity, enhanced mineralization capacity. obtained opposite trends. MiRNA-27a-3p could target ATF3 reversed promotive on differentiation.MiRNA-27a-3p promotes into osteoblasts symptoms.
