Effect of Carbidopa on 18F-FDOPA Uptake in Insulinoma: From Cell Culture to Small-Animal PET Imaging

ABSTRACT
Patient premedication with carbidopa seems to improve the accuracy of 6-18F-fluoro-3,4-dihydroxy-l-phenylalanine (18F-FDOPA) PET for insulinoma diagnosis. However, risk false-negative results in presence is a concern. Consequently, we aimed evaluate effect on 18F-FDOPA uptake β-cells and an xenograft model mice.18F-FDOPA vitro accumulation was assessed murine β-cell line RIN-m5F. In vivo small-animal experiments were performed tumor-bearing nude mice after subcutaneous injection RIN-m5F cells. Experiments conducted without pretreatment.Incubation cells 80 μM did not significantly affect cellular 18F-FDOPA. Tumor xenografts clearly detectable by all cases. Insulinoma carbidopa-treated showed higher than those nontreated mice. Regardless premedication, characterized early increase then progressive reduction over time.Carbidopa influence but improved imaging vivo. Our findings current knowledge about profile related model. To our knowledge, present work represents first preclinical research specifically focused insulinomas, potential translational implications.
