ABSTRACT
Stroke has been the leading cause of adult morbidity and mortality over past several years. After an ischemic stroke attack, many dormant or reversibly injured brain cells exist in penumbra area. However, pathological processes unique cell information area acute remain elusive. We applied unbiased single sequencing combination with bulk RNA-seq analysis to investigate heterogeneity each type early stages detect possible therapeutic targets help survival. used these analyses study mouse during this phase. Our results reveal impact on specific genes pathways different types alterations differentiation trajectories, suggesting potential mechanisms targets. In addition classical gene markers, single-cell genomics demonstrates subclusters metabolism changes stroke. These findings suggest that Gadd45b microglia, Cyr61 astrocytes, Sgk3 oligodendrocytes may play a subcluster-specific role death survival Moreover, RNA-scope multiplex situ hybridization immunofluorescence staining were selected target markers validate confirm existence subtypes molecular stage
