Identification of testis development-related genes by combining Iso-Seq and RNA-Seq in Zeugodacus tau

ABSTRACT
Introduction: Zeugodacus tau (Walker) is an invasive pest. An effective method to control this pest the sterile insect technique (SIT). To better apply technique, it necessary understand testis development progression. Methods: Differentially expressed genes (DEGs) during were analyzed by PacBio Iso-Seq and RNA-seq. Results: RNA-Seq library of Z. testes on day 1, 6, 11 post eclosion constructed. We identified 755 865 differentially in comparisons T6 (testes 6) vs. T1 T11 T1, respectively. The KEGG pathway analysis showed that DEGs significantly enriched retinol metabolism, vitamin B6 ascorbate aldarate metabolism pathways. Knockdown dehydrogenase 12 -like ( rdh12 -like), pyridoxal kinase (pdxk) regucalcin (rgn) , representative gene each above 3 pathways, reduced hatching rate offspring. In addition, we 107 Drosophila spermatogenesis-related orthologous which innexin 2 (inx2) exhibited up-regulated expression throughout development, knockdown offspring rate. Discussion: Our data indicated -like, pdxk rgn inx2 related they conserved tephritid species. These results suggested might have same function tephritid. findings provide insight into spermatogenesis
