ABSTRACT
Septic cardiomyopathy (SCM) is a serious complication caused by sepsis that will further exacerbate the patient's prognosis. However, immune-related genes (IRGs) and their molecular mechanism during septic are largely unknown. Therefore, our study aims to explore hub (IRHGs) miRNA-mRNA pairs with potential biological regulation in SCM means of bioinformatics analysis experimental validation.Firstly, screen differentially expressed mRNAs (DE-mRNAs) from dataset GSE79962, construct PPI network DE-mRNAs. Secondly, were identified overlapped immune cell marker (ICMGs) obtain IRHGs SCM. In addition, receiver operating characteristic (ROC) curve was also performed this process determine disease diagnostic capability IRHGs. Finally, crucial miRNA-IRHG regulatory predicted constructed bioinformatic methods. Real-time quantitative reverse transcription-PCR (qRT-PCR) GSE72380 used validate expression key axis.The results infiltration showed neutrophils, Th17 cells, Tfh central memory cells had more than control group; A total 2 obtained crossing gene ICMGs, validated qRT-PCR. Ultimately, we IRHG SCM: THBS1. The ROC THBS1 area under (AUC) 0.909. miR-222-3p/THBS1 axis constructed.In summary, propose may be IRHG, can serve as biomarker for diagnosis SCM; involved pathogenesis promising candidate therapy.
