ABSTRACT
Growth differentiation factor 15 (GDF15) is involved in the occurrence and development of many diseases, there are few studies on its relationship with sepsis. This article aims to explore clinical value GDF15 sepsis preliminarily prospective regulatory effect macrophage inflammation functions. We recruited 320 subjects (132 cases group, 93 nonsepsis 95 control group), then detected serum levels laboratory indicators, further investigated correlation between also analyzed diagnosis, severity assessment, prognosis. In vitro , we used LPS stimulate THP-1 RAW264.7 cells establish inflammatory model, expression culture medium under state. After that, added recombinant protein (rGDF15) pretreatment The results showed that were significantly increased which was correlated indexes organ damage, coagulation indexes, factors, SOFA score. has a high AUC diagnosis sepsis, can be improved by combining other indicators. dynamic monitoring play an important role judgment prognosis state, intracellular extracellular increased. reduce cytokines, inhibit M1 polarization induced LPS, promote M2 polarization. Moreover, enhances phagocytosis bactericidal function macrophages. Finally, observed decreased level phosphorylation JAK1/STAT3 signaling pathway nuclear translocation NF-κB p65 rGDF15. summary, our study found good application plays protective regulating functions macrophages inhibiting activation p65.
