Nanopore-based targeted next-generation sequencing of tissue samples for tuberculosis diagnosis

ABSTRACT
Objective Diagnosing tuberculosis (TB) can be particularly challenging in the absence of sputum for pulmonary cases and extrapulmonary TB (EPTB). This study evaluated utility nanopore-based targeted next-generation sequencing (tNGS) diagnosing tissue samples, compared its efficacy with other established diagnostic methods. Methods A total 110 samples from clinical were examined. The sensitivity specificity tNGS benchmarked against a range existing approaches including hematoxylin eosin (HE) staining conjunction acid-fast bacilli (AFB) detection, HE combined PCR, paired immunohistochemistry (IHC) using anti-MPT64, Xpert Mycobacterium (MTB)/rifampicin (RIF) assay. Results 88.2 94.1%, respectively. respective sensitivities AFB, IHC MTB/RIF 30.1, 49.5, 47.3, 59.1%. specificities these methods 82.4, 88.2, 94.1, Analysis drug resistance based on results indicated that 10 93 patients (10.75%) had potential resistance. Conclusion Targeted achieved higher accuracy than methods, play crucial role rapid accurate diagnosis TB, drug-resistant TB.
