Mitochondrial membrane models built from native lipid extracts: Interfacial and transport properties

ABSTRACT
The mitochondrion is an essential organelle enclosed by two membranes whose functionalities depend on their very specific protein and lipid compositions. Proteins from the outer mitochondrial membrane (OMM) are specialized in dynamics mitophagy, whereas proteins of inner (IMM) have dedicated functions cellular respiration apoptosis. As for lipids, OMM enriched glycerophosphatidyl choline but cardiolipin exclusively found within IMM. Though topology distribution IMM known since more than four decades, little about interfacial dynamic properties extracts. Here we build monolayers, supported bilayers giant unilamellar vesicles (GUVs) native lipids extracts porcine heart. Additionally, perform a comparative analysis interfacial, phase immiscibility mechanical both types extract. Our results show that form expanded softer allowing better understanding physicochemical biophysical membranes.
