DJ-1 promotes osteosarcoma progression through activating CDK4/RB/E2F1 signaling pathway

ABSTRACT
Osteosarcoma (OS) is a primary malignant tumor of the bone characterized by poor prognosis due to chemotherapy resistance and high recurrence rates. DJ-1 (PARK7) known as an oncogene its abnormal expression related various types tumors. It was found in this study that upregulated closely correlated with OS patients promoting proliferation, migration cells vitro through regulating activity CDK4 but not oxidation mechanism or AKT pathway. The combination promoted RB phosphorylation, leading dissociation E2F1 into nucleus regulate cell cycle-related genes. xenograft mouse model demonstrated knockout suppressed growth vivo. All these findings indicate can affect occurrence progression CDK/RB/E2F1axis, suggesting novel therapeutic opportunity for patients.
