ABSTRACT
Diabetic cardiomyopathy (DCM) is a serious complication of diabetes that can lead to heart failure and death, for which there no effective treatment. Rhynchophylline (Rhy) the main component Chinese herbal medicine Uncaria rhynchophylla , mainly acts on cardiovascular nervous systems. However, its role in protecting against DCM remains unexplored. The present study sought reveal mechanism Rhy improving type 2 mellitus (T2DM) myocardial lesions from perspective regulating calcium homeostasis cardiomyocytes. We prepared mouse model T2DM using high-fat diet combined with low doses streptozotocin. mice were given 40 mg/kg 8 weeks. results showed attenuate cardiac pathological changes, slow down rate, decrease serum enzyme levels, reduce cardiomyocyte apoptosis, enhance contractility, raise transient amplitude mice. Further, downregulated phosphorylation level ryanodine receptor 2, upregulated phospholamban, protected mitochondrial structure function, increased adenosine triphosphate levels tissue Our demonstrated may protect damage promote contraction, action seems be related regulation intracellular homeostasis.
