ABSTRACT
Myocardial infarction (MI) is a highly prevalent and lethal disease worldwide. Prevention timely recovery are critical for the control of recurrence heart failure in MI survivors. The present study was designed to investigate cardioprotective activity herbal medicine formula Baoyuan Decoction (BYD) identify active compounds molecular targets. ethanolic BYD extract (BYDE) prepared by water extraction ethanol precipitation four medicines, Astragali Radix, Ginseng Radix et Rhizoma, Cinnamomi Cortex, Glycyrrhizae Rhizoma. Initially, BYDE validated effectiveness mouse model ischemia injury rat cardiomyocyte H9C2 cells. As results, effectively reduced infarct size from 56% 37% preserved cardiac functions while protected cells against oxygen glucose deprivation. Subsequent network pharmacology analysis revealed that 122 bioactive ingredients, including flavonoids saponins UPLC-MS/MS profile BYDE, might target 37 MI-related proteins, inflammatory apoptotic mediators (e.g., TNF, NFKB1, CASPs, TNFRSF1A, CXCL12, BCL2A1). Pathway enrichment suggested inflammation via targeting tumor necrosis factor-alpha (TNF-α)/nuclear factor-κB (NF-κB) pathway selected targets were also implicated IL-17 signaling pathway, lipid atherosclerosis. Consequently, adenosine, ginsenoside Rh2, isoliquiritigenin, licochalcone A generate four-compound mixture AGILe inhibitory effects on TNF-α/NF-κB pathway. results be potential remedy MI.
