ABSTRACT
To investigate whether the abscopal effects of cranial irradiation (C-irradiation) cause testicular damage in mice, male C57BL/6 mice (9weeks age) were randomly divided into a sham group, shielded group and C-irradiation administered sham/shielded or at dose rate 2.33Gy/min (5Gy/d for 4 d consecutively). All sacrificed 4weeks after C-irradiation. We calculated testis index, observed histology by haematoxylin-eosin (HE) staining ultrastructure transmission electron microscopy. Western blotting was used to determine protein levels Bax, Bcl-2, Cleaved caspase 3, glial cell line-derived neurotrophic factor (GDNF) stem (SCF) testes mice. Immunofluorescence performed detect expression 3 3β hydroxysteroid dehydrogenase (3βHSD), TUNEL assay confirm location apoptotic cells. The testosterone (T), GDNF SCF measured ELISA. also evaluated sperm quality cauda epididymides measuring count, abnormality, survival apoptosis rate. results showed that there no significant difference histology, between group. Compared with exhibited lower index severely damaged increased markedly especially spermatogonial serum T 3βHSD did not obviously differ but compared In addition, count decreased while abnormality increased. Under these experimental conditions, induced regard both structure function ultimately These findings provide novel insights prevention treatment targets reproductive
