Identification of Circular RNA Expression Profiles in White Adipocytes and Their Roles in Adipogenesis

ABSTRACT
Obesity and its related metabolic diseases have become great public health threats worldwide. Although accumulated evidence suggests that circRNA is a new type of non-coding RNAs regulating various physiological pathological processes, little attention has been paid to the expression profiles functions circRNAs in white adipose tissue. In this study, 3,771 were detected three stages adipogenesis (preadipocyte, differentiating preadipocyte, mature adipocyte) by RNA-seq. Experimental validation suggested RNA-seq results are highly reliable. We found nearly 10% genes which expressed linear adipocytes could also generate circRNAs. addition, 40% them produced multiple isoforms. performed correlation analysis deal (nearly 50%) their parental correlated levels. A total 41 differential (DECs) during an extremely high ratio (80%) with genes, indicating these may potentially play roles genes. KEGG enrichment GO annotation suggesting DECs participate several adipogenesis-related pathways. Following rigorous selection, we many up-regulated contain miRNAs binding sites, such as miR17, miR-30c, miR-130, they facilitate regulatory acting miRNA sponges. These suggest plenty serve candidates for future regulation.
