Electroacupuncture pre-treatment alleviates sepsis-induced cardiac inflammation and dysfunction by inhibiting the calpain-2/STAT3 pathway

ABSTRACT
Electroacupuncture (EA) has both anti-inflammatory and cardio-protective effects. Activation of calpain pathway is involved in several myocardiopathy. In sepsis, the role calpain-2-regulated STAT3 mechanism electroacupuncture remains unclear. this study, we aimed to elucidate by which reduces cardiac inflammation apoptosis improves function during sepsis. pretreatment for 7 days was applied septic cardiomyopathy model induced lipopolysaccharide (LPS). lipopolysaccharide-induced sepsis associated with a dramatically systemic dysfunction, alleviated pre-treatment. Lipopolysaccharide resulted increases pro-inflammatory factors (TNF-α,IL1βand IL-6) (TUNEL staining BAX/Bcl2) via activation calpain-2/STAT3 pathway.Electroacupuncture pre-treatment inhibited LPS-induced signalling ameliorated inflammatory apoptosis. Additionally, inhibition calpain-2 expression using corresponding siRNA decreased Phosphorylation STAT3,pro-inflammatory lipopolysaccharide- treated cardiomyocytes, confirming that activated p-STAT3 participate cardiomyopathy. Furthermore, suppression stattic enhanced anti-apoptosis effects electroacupuncture. These findings reveal mechanisms preconditioning protection against mouse may provide novel targets clinical treatments sepsis-induced dysfunction.
