ABSTRACT
Genetic transformation is an important strategy for revealing gene function, and it used extensively in both functional genomics study molecular breeding of rice. Demand its application wild Oryza species rising their extensive genetic diversity. However, accessions with AA genome using calli induced from scutellum tissue embryos mature seeds has not been successfully established. In the present study, we Chaling common rice (CLCWR) (Oryza rufipogon Griff.) to establish Agrobacterium-mediated system based on seeds. The CLCWR were easy be regenerated. callus induction rate texture optimum under 2.5 mg/L 2,4-D. optimal hormone combination regeneration was 2 ZT + 0.1 NAA. Studies editing showed that efficiency 87-94%, single multiplex about 60-70% 20-40%, respectively. Compared Nipponbare (Nip), had higher Hygromycin-resistant frequency efficiency. Taken together, our establishes a highly efficient provides good material de novo domestication by future.
