ABSTRACT
Powdery mildew (PM) is one of the main fungal diseases that appear during cultivation melon fruit crop. Mildew Resistance Locus "O" (MLO) known as a gene family and has seven conserved transmembrane domains. An induced functional loss specific MLO could mainly confer PM resistance to melons. However, genomic structure genes its role in still remain unclear melon. In this study, bioinformatic analysis identified total 14 members genome sequence, these were distributed an uneven manner on eight chromosomes. The phylogenetic divided CmMLO into five different clades, structural showed same clade had similar intron exon distribution patterns. addition, by cloning sequence four lines, analyzing expression pattern after infection, making microscopic observations infection PM, we concluded CmMLO5 (MELO3C012438) plays negative regulating PM-resistance susceptible line (Topmark), critical time point for function was noticed at 24 72 hours infection. mutational exhibited single base mutation 572 bp, which further results protein function, thus conferring summary, our research evidence provides thorough understanding demonstrates their potential disease resistance, well theoretical foundation breeding.
