ABSTRACT
Stilbenes, particularly trans -resveratrol, play a highly relevant defense role in grapevines as phytoalexin is induced response to stress. Metabolism and transport of stilbenes can be conveniently investigated grapevine cell culture since large amounts -resveratrol are accumulated the extracellular medium upon treatment with elicitor methylated cyclodextrin, either alone or combined methyl jasmonate. A proteomic approach on membrane fractions was performed find transporter candidates. The candidate VvABCB15 functionally characterized. Its stable expression both yeast Silybum marianum cells’ heterologous systems led increased these hosts. Transient Vitis cells showed an enhanced absorbent- elicitor-assisted accumulation VvABCB15-expressing VvGSTU10/VvABCB15-co-expressing suspension cultures. Experiments transient suspensions using light-switchable stilbene synthase (pHYH::VvSTS3) further confirmed candidate’s transporter. VvABCB15-YFP fusion proteins Nicotiana leaf localization plasma membrane, consistent functional transport. This first report provide evidence for involvement ABC B type, VvABCB15, cells.
