ABSTRACT
Arginine has a positive effect on pre-implantation development in pigs. However, the exact mechanism by which arginine promotes embryonic is undefined. Here, single-cell RNA sequencing technology was applied to porcine vivo embryos from zygote morula stage, it found that expression of metabolism-related genes clearly changed 2-cell stage 4-cell when zygotic genome activation (ZGA) occurs embryos. Further analysis showed are significantly correlated with key ZGA genes. To determine function during ZGA, vitro fertilization were cultured PZM-3 medium (0.12 mM arginine, Control group), modified (0 Block group) and supplemented + Arg group). The results arrest rate increased group compared (P < 0.05). decreased than Meanwhile, marker SIRT1 protein group, their group. In addition, we observed glutathione (GSH), ATP levels, lipid droplet contents increased, reactive oxygen species (ROS) level Compared spermine content culture mRNA ornithine decarboxylase1 (ODC1) 0.05), those Moreover, difluoromethylornithine (an inhibitor ODC1) added inhibited. summary, our findings demonstrated may regulate under nutrition restriction promoting polyamine synthesis.
