ABSTRACT
In a series of previous studies, we demonstrated that the photodynamic therapy (PDT), as widely used tool for treatment glioblastoma multiforme (GBM), also site-specifically opens blood–brain barrier (BBB) in PDT-dose and age-related manner via reversible disorganization tight junction machinery. To develop effective protocol PDT-opening BBB, here answer question what kind photosensitizer (PS) is most BBB opening. We studied healthy mice using commercial photosensitizers (PSs) such 5-aminolevulenic acid (5-ALA), aluminum phthalocyanine disulfonate (AlPcS), zinc (ZnPc) new synthetized PSs galactose functionalized ZnPc (GalZnPc). The spectrofluorimetric assay Evans Blue albumin complex (EBAC) leakage 3-D confocal imaging FITC-dextran 70 kDa (FITCD) extravasation clearly shows revisable dose depended to EBAC FITCD associated with decrease presence (TJ) vascular endothelium. PDT effects on permeability, TJ expression fluorescent signal from brain tissues are more pronounced PDT-GalZnPc vs. PDT-5-ALA/AlPcS/ZnPc. These pre-clinical data first important informative platform an optimization light knowledge about drug delivery diseases.
