ABSTRACT
Metastasis Associated Lung Adenocarcinoma Transcript-1 (MALAT1) is implicated in regulating the inflammatory response and pathology of several chronic diseases, including osteoarthritis (OA). The purpose this study was to examine relationship between OA subchondral bone expression MALAT1 with parameters joint health biomarkers inflammation, determine its functional role human osteoblasts. Subchondral blood were collected from hip knee patients (n = 17) only neck femur fracture 6) undergoing replacement surgery. Cytokines determined by multiplex assays ELISA, gene qPCR. loss function performed patient osteoblasts using locked nucleic acids. osteoblast transcriptome analysed RNASeq pathway analysis. Bone positively correlated serum DKK1 galectin-1 concentrations, induced IL-1β stimulation. Osteoblasts depleted exhibited differential (>1.5 fold change) 155 genes, PTGS2. Both basal IL-1β-mediated PGE2 secretion greater induction upon challenge modulation production suggests that may play a inflammation bone.
