ABSTRACT
Peroxidasin (PXDN), a human homolog of Drosophila PXDN, belongs to the family heme peroxidases and has been found promote oxidative stress in cardiovascular tissue, however, its role prostate cancer not previously elucidated. We hypothesized that PXDN promotes progression via regulation metabolic pathways. analyzed expression tissue by immunohistochemistry increased with as compared normal or cells. knockdown followed proteomic analysis revealed an increase stress, mitochondrial dysfunction gluconeogenesis Additionally, Liquid Chromatography tandem mass spectrometry (LC-MS/MS)-based metabolomics confirmed induced global reprogramming associated decreased nucleotide biosynthesis. further demonstrated led reactive oxygen species (ROS) cell viability apoptosis. Finally, colony formation on soft agar. Overall, data suggest regulating metabolome, more specifically, inhibiting leading Therefore, may be biomarker potential therapeutic target.
