ABSTRACT
Introduction:Recent studies have drawn attention to the use of gold nanoparticles (AuNPs) in immunobiological production.Ongoing efforts effective production peptide-derived monoclonal antibodies (mAbs) shown trend developing target-specific systems, which antigen is conjugated AuNPs for targeted delivery antigen-presenting cells (APCs).Several describe them as promising vehicles immunotherapy diseases such cancer, due their ability stimulate innate immunity.Thus, they play an interesting dual role, acting adjuvant and carrier antigen, being able reduce toxicity system increase its immunogenic activity, promoting significant antigen-specific responses, without need traditional adjuvants.Objective: Thus , study aims AuNps carriers peptides adjuvants initial platform anti-cancer antibodies.Methodology: Based on this, a suitable medium was prepared exposure binding peptides, previously produced murine immune response induction.For functionalizations 900μl contact with 100μl colorectal cancer marker were performed, under constant magnetic stirring, 24 hours before each inoculation.After functionalization, BALB/c mice immunized nanosystem ten doses at 15-day intervals each.Serum samples obtained after immunization subjected ELISA (Enzyme-Linked Immunosorbent Assay) evaluate response, control group only peptide 0.01% PBS.The spleen macerated nylon compression obtain B lymphocytes, fused myeloma hybridomas.These cultured selective HAT (hypoxanthine-aminopterin-thiamine) supernatant submitted determine wells containing polyclonal antibody producing mAbs purified two steps positive clone expanded largescale production. Results:The final product concentrated yield equal 1.3mg/mL pAbs 1.14mg/ mL mAbs. Conclusion:The demonstrated efficiency proposed immunobiological, since several challenges encountered nano-system, preventing through alone.This project serves basis further using AuNps/Peptides based nanosystems primary platform.
