ABSTRACT
Porcine circovirus type 2 (PCV2) is one of the most significant pathogens in global pig industry. Due to fact that distinctive feature porcine circovirus-associated diseases (PCVAD) a chronic course, it necessary introduce effective serodiagnostics methods determine viral kinetics populations. The aim study was develop an indirect enzyme-linked immunosorbent assay (ELISA) based on recombinant capsid protein detect antibodies PCV2. At first stage, structure analyzed, which dominant regions for antibody recognition were determined. These fragments included multiepitope insert encoding target diagnostic antigen with molecular weight 17,8 kDa. As result expression ORF-2 gene fragment E. coli cells, soluble obtained and purified electrophoretically homogeneous state using metal chelate chromatography. Using this antigen, taking into account recommendations World Organization Animal Health (WOAH), conditions ELISA optimized. A preliminary assessment analytical characteristics carried out sample known positive negative blood sera pigs, as well 181 pigs aged 3 9 months, collected from different farms Republic Tatarstan. It found average seroprevalence PCV2 individual populations 72,9 % (95% CI: 66,0–78,9 %). However, dynamics PCVAD populations, further serological monitoring combination virological profile herd.
