ABSTRACT
Objective
To evaluate the influence of preconditioning with and anti-myosin-monoclonal antibody (mAb2G4)-nuclear factor-kappa B decoy oligodeoxynucleotide (ODN)-lipofectamine (lip) on hypoxia-reoxygenation (H/R) injury in H9c2 cardiomyocytes.
Methods
H9c2 cardiomyocytes were seeded 6-well plate at density 1×105/ml (2 ml/well), divided into 3 groups (n=9 each) using a random number table: control group (group C), H/R mAb2G4-ODN-lip MOL). The cells underwent 2 h hypoxia an air-tight bag, followed by 1 reoxygenation. In MOL group, treated μg ODN) for 4 then cultured common culture medium 8 before hypoxia. At end reoxygenation, proliferation was measured MTT assay, supernatant collected to determine activity lactate dehydrogenas (LDH), content malondialdehyde (MDA), concentrations tumor necrosis factor-α (TNF-α) interleukin-6 (IL-6) (by ELISA). rate inhibition calculated.
Results
Compared C, inhibition, LDH activity, MDA content, TNF-α IL-6 significantly increased other two groups. Compared H/R, decreased group.
Conclusion
mAb2G4-ODN-lip can mitigate cardiomyocytes.
