Effect of bone marrow mesenchymal stem cells on liver fibrosis of rats

ABSTRACT
Objective To observe bone mesenchymal stem cells (BMSCs) influence on the development of liver fibrosis.Methods We choose 20 female Wistar rats which randomly divided into intervention group (10 rats) and control rats).Interventional were accepted water involve consistency as 0.01% diethylnitrosamine.In 4,8,12 week we transplant 1 × 106 BMSCs suspension male rat to interventional through caudal vein.Control injected equal normal saline vein.The breed in same condition for 21 weeks observed following index:(1) By hematoxylin eosin (HE) staining,observe pathomorphological change liver.(2) Detect existence SrY by hybridization situ confirm successful migrate BMSCs.(3) VG special coloration detect expression level α-smooth muscle actin (α-SMA) immunohistochemistry differences two groups.Results There are 7 9 have developed fibrosis HE staining; discover group.the positive rate α-SMA was (21.97 ± 11.00)% ; higher than (10.30 3.36)%,there is significant difference (t =2.69,P < 0.05).The (20.63 10.02) % apparently (11.24 3.80) %,there (F =2.34,P <0.05).Conclusion In model fibrosis,BMSCs has function promote rat.
