The protective effect of JNK inhibitor SP600125 on hippocampal neurons in rats with status epilepsy

ABSTRACT
Objective To explore the protective effect and probable mechanism of JNK inhibitor SP600125 on hippocampal neurons in rats with status epilepsy following lithium-pilocarpine. Methods 48 Wistar rats, accordance random number table, were divided into control, epilepticus (SE) group (SP). HE staining fluorescent TUNEL method used to observe pathological changes neuronal apoptosis area each group. Western blot was applied detect phosphorylation expression its downstream effector molecule c-JUN tissues group. Results Compared control group, loss CA3 hippocampus SE significant (percentage positive cells (26.34±3.04)%, P<0.05). The mortality significantly decreased obviously reduced SP than (mortality : 6.25%, 37.5% respectively, Meanwhile, levels phospho-JNK phospho-c-JUN increased (The relative OD values respectively 0.447±0.025, 0.552±0.035, P<0.05 compared Control group). After treated 0.211±0.016, 0.237±0.028, group). Conclusion JNK may play an important rat after through inhibition phosphorylation.
