Anti-cicatricial effect of tetrandrine drug delivery system in glaucoma filtration surgery in rabbit

ABSTRACT
Background Scarring of the filtering bleb is a main cause surgical failure in glaucoma.It has been reposed that tetrandrine could suppress proliferation cultured human fibroblast Tenons capsule vitro and thus potential effect to prevent scarring after surgery. Objective Present study was investigate anti-cicatricial drug delivery system(Tet DDS)during filtration Methods Filtration surgery performed bilateral eyes 18 New Zealand white rabbits.The Tet DDS with 0.3 mg Tet,0.2 or free-Tet were implanted subcunjunctially during surgery.The blebs scored 1 day,4,7,10,14 days referring corneal thickness range under slit-lamp biomicroscopy.The morphology assessed by vivo confocal microscopy 7 14 operation.The specimen prepared for histopathological examination. Results The scores implantation groups significantly higher than those group from 4 through trabeculectomy(P<0.01),and showed considerably increase compared 0.2 trabeculectomy(P<0.05).The found distinct subepithelial cystic spaces light on 7th day 14th surgery.Compared group,the numbers mierocysts much more(P<0.01)and area microcysts larger(P<0.01)in group.The tissue presented more larger operation(P<0.05).The inflammatory cell infiltration wag milder comparison free-Ted group.Conclusion strong inhibitory effects cells activity fibroblagt early stage therefore improve success rate.
