ABSTRACT
Cell death is a fundamental process in all living organisms. The protocol establishes lipopolysaccharide (LPS) and adenosine triphosphate (ATP)-induced phorbol-12-myristate-13-acetate (PMA)-differentiated lipid deposition human monocyte (THP-1) macrophage model to observe cell death. LPS combined with ATP classic inflammatory induction method, often used study pyroptosis, but apoptosis necroptosis also respond stimulation by LPS/ATP. Under normal circumstances, phosphatidylserine only localized the inner leaflet of plasma membrane. However, early stages apoptosis, necroptosis, membrane remains intact exposed phosphatidylserine, later stages, loses its integrity. Here, flow cytometry was analyze Annexin V 7-Aminoactinomycin D (AAD) double staining detect from whole cells. results show that substantial cells died after Using scanning electron microscopy, we possible forms individual indicate may undergo or This focuses on observing macrophages showed not limited pyroptosis necrotic can occur, helping researchers better understand choose experimental method.
