ABSTRACT
Radix platycodi is the root of Platycodon grandiflorus A. DC, which has been widely used as a food material and for treatment number chronic inflammatory diseases in traditional oriental medicine. In this study, anti‑inflammatory effects saponins isolated from radix (PGS) on production mediators cytokines lipopolysaccharide (LPS)-stimulated BV2 murine microglial cells were examined. We also investigated PGS LPS‑induced nuclear factor‑κB (NF-κB) activation phosphoinositide 3-kinase (PI3K)/AKT mitogen-activated protein kinase (MAPK) signaling pathways. Following stimulation with LPS, elevated nitric oxide (NO), prostaglandin E2 (PGE2) pro-inflammatory cytokine was detected cells. However, significantly inhibited excessive NO, PGE2 pro‑inflammatory cytokines, including interleukin-1β (IL-1β) tumor necrosis factor-α (TNF-α) concentration-dependent manner without causing any cytotoxic effects. addition, suppressed NF-κB translocation LPS-induced phosphorylation AKT MAPKs. Our results indicate that inhibitory effect LPS-stimulated response associated suppression PI3K/AKT MAPK Therefore, these findings suggest may be useful neurodegenerative by inhibiting responses activated
