ABSTRACT
There are literally thousands of known agents with potential chemopreventive and antioxidant activity; however, the expanding list natural synthetic compounds makes it difficult to test every agent in widely accepted 2-year animal bioassay human clinical trials. Therefore, short-term screening assays needed sort out most efficacious for long-term studies. In present study, identification was explored a Cu2+-mediated Fenton-type reaction, coupled oxidative DNA lesion detection by 32P-postlabeling. Several inhibited formation 8-oxo-2'-deoxyguanosine (8-oxodG), benchmark lesion, but ellagic acid, polyphenol found berries, offered maximal (>80%) inhibition 8-oxodG formation. However, well-known tea polyphenol, epigallocatechin gallate, along silymarin D,L-sulforaphane, exhibited pro-oxidant effect, 50-70% increase induction. general, our results agree reported - activities compounds, rendering this vitro assay be useful determining rapidly cost-effectively.
