ABSTRACT
Ischemic stroke, the third leading cause of disability globally, imposes a notable economic burden. Tetrandrine (Tet), which has been widely used clinically, exhibits potential protective effects against stroke. However, there little pre‑clinical research to evaluate therapeutic Tet on The present study investigated beneficial effect ischemia‑reperfusion (I/R) injury and its underlying mechanism in rats. Rats were subjected occlusion middle cerebral artery, then treated with (30 mg/kg/day, intraperitoneal) subacute phase for 7 days. In order detect behavior rats, modified neurological severity score longa behavior, grasping capability inclined plane tests conducted days 1, 3 following ischemia. addition, neuronal apoptosis cortex hippocampus ischemia was assessed by Nissl staining TUNEL assay. Finally, oxidative stress evaluated measurement free radicals immunofluorescence LC3 assess autophagy. improved function decreased infarct volume I/R also prevented region. protected damage ischemia, reflected levels nitric oxide malondialdehyde increased glutathione (GSH) GSH peroxidase. expression autophagy marker treatment group. conclusion, attenuated I/R‑induced decreasing stress,
