ABSTRACT
Cluster of differentiation 47 (CD47) is a transmembrane protein that widely and moderately expressed on the surface various cells can have an essential role in mediating cell proliferation, migration, phagocytosis, apoptosis, immune homeostasis other related responses by binding to its ligands, integrins, thrombospondin‑1 signal regulatory α. The poor prognosis cancer patients closely associated with high expression CD47 glioblastoma, ovarian cancer, breast bladder colon hepatocellular carcinoma. Upregulation facilitates growth numerous types tumor cells, while downregulation promotes phagocytosis macrophages, thereby limiting growth. In addition, blocking activates cyclic GMP‑AMP (cGAMP) synthase/cGAMP/interferon gene stimulating factor signaling pathway initiates adaptive response kills cells. present review describes structure, function interactions as well regulation fate. It summarizes therapeutics, mechanisms action, research advances challenges targeting CD47. this paper provides overview latest therapeutic options for CD47, such chimeric antigen receptor (CAR) T‑cells, CAR macrophages nanotechnology‑based delivery systems, which are future clinical
