ABSTRACT
Abstract Mast cells and basophils, which are activated by IgE allergens through the high affinity receptor (Fc epsilon RI), play a prominent role in anaphylaxis mouse. Mice deficient this become resistant to passive anaphylaxis. As first step developing an vivo model that more closely mimics IgE-mediated responses man, we used combination of transgenic embryonic stem cell technology generate mouse line murine Fc RI alpha-chain has been replaced with its human homologue. We demonstrate here these mice express tetrameric receptor, associates beta- gamma-chains, upon triggering relevant Ag, mediates initiation expected intracellular events. In addition, show restores anaphylactic response nonresponsive alpha-deficient parental line. This "humanized" represents potentially important system, not only for studying immune responses, but also testing potential therapeutic reagents can interfere mediated receptor.
