ABSTRACT
Abstract The mainstay of immunosuppression is Prograf (FK), Cyclosporine (CSA), and Sirolimus (SRL). These drugs target T cells which are pivotal in transplant rejection. Little known about their effects on length cell suppression once stopped. We hypothesized that reactivity would return later with SRL compared to CSA or FK. 18 health volunteers were recruited. 8 received FK, 3 CSA, 7 SRL. FK taken twice daily for 3.5 days 4 days. Drugs titrated respective troughs (FK 8-10 mg/L, 150-200 mcg/L, 10-15 ng/ml). Blood drawn at 0, 12, 24, & 48 hrs after last dose. CD4 isolated using isolation beads, stimulated CD3/28 cultured 96 well plates. Cell proliferation ATP concentration measured BrdU labeling Cylex assay respectively. At 24 hrs, fold change between by was not statistically significant (1.0 vs. 0.8, p=0.2). time 0 2/8 patients 12 4/8 group, 1/3 6/7 the group (p<0.05). Proportions but did trend towards significance (p=0.07). In conclusion, associated longer in-vitro taking CSA.
