Comprehensive profiling of TCR and BCR repertoire of infiltrated lymphocytes in the mouse injured skeletal muscle

ABSTRACT
Abstract Lymphocytes infiltrating to the injured skeletal muscle have been shown play an important role in regeneration. Site-specific infiltrating/expanding lymphocytes and its molecular features are potential resources for bioengineering such as a drug delivery system. Although some previous studies showed accumulation of muscle, actual frequency distinct lymphocyte clones situ is still unclear. Hence, comprehensive picture repertoire has yet be described detail. To this end, we conducted profiling TCR BCR mouse model by next-generation sequencing. In profiling, found that expression level transcripts contains unique CDR3 α population β were increased (7.6–17% 6.7–15%) T cells both cardiotoxin- or cryo-injured whereas no clonal expansion specific α/β observed control splenic (<1% respectively). While various immunoglobulin kappa chains infiltrated fraction, despite proportion heavy lambda chain remained similar B cells. Together these data allowed us identify TCR/BCR candidates targeting muscles. Furthermore, aim utilize cells/B expressing high-affinity system, especially large protein complexes.
