Analysis of Gluten Proteins Composition During Grain Filling in Two Durum Wheat Cultivars Submitted to Two Water Regimes

ABSTRACT
Durum wheat (Triticum turgidum L. subsp. durum) is one of the major crops in Mediterranean basin, where water stress often occurs during grain filling which represents a critical stage for synthesis and accumulation storage proteins (gliadins glutenins). The aim study to evaluate, by two-dimensional gel electrophoresis (2DE SDS-PAGE), composition two durum cultivars (Ciccio Svevo) cultivated growth chamber under different regimes (control deficit). At milk physiological maturity, gluten have been extracted separated 2DE SDS-PAGE. analysis gels was performed software ImageMaster 2D Platinum (Amersham). results showed differences protein expression within regions between cultivars; deficit rate faster all regions, either at stage. Protein high molecular weight (H) region resulted Ciccio than Svevo mainly control treatment. In low 48 35 kDa (L 48-35), cultivar higher Svevo. Furthermore marked increase H volume decrease L 48-35 observed only Svevo; instead no change showing this greater stability on changing regime. Further studies use mass spectrometry are necessary identify specific peptides relative drought as well investigate relationships with technological quality.
