ABSTRACT
Forensic odontology necessarily involves the application of dentistry along with various other branches sciences which deals proper handling, examination, evaluation, and presentation dental evidences, that aids to investigate a crime deliver justice. Sex determination is part forensic an essential priority when traditional identification deceased becomes impossible. To determine by analysis Amelogenin gene using Polymerase Chain Reaction (PCR) method on Deoxyribose nucleic acid (DNA) isolated from pulp, was exposed environmental conditions created artificially mimic scenario. This in-vitro study conducted subjecting extracted teeth imitating scene, viz. desiccation at room temperatures, immersion in salt water, burial soil even exposing extremes temperatures. DNA pulp tissue sex achieved amplification amelogenin through AMEL based primers PCR.Among all samples used this study, could be all, except those were subjected temperature 350 °C. found accurate compared individual recorded initially, during collection samples. This shows potent source extreme conditions, barring high temperatures PCR markers quite reliable.
