Synergistic Tumoricidal Effects of Alpha-Lipoic Acid and Radiotherapy on Human Breast Cancer Cells via HMGB1

ABSTRACT
Radiotherapy (RT) is one of main strategies cancer treatment. However, some cells are resistant to radiation-induced cell death, including apoptosis. Therefore, alternative approaches targeting different anti-tumor mechanisms such as senescence required. This study aimed investigate the synergistic effect alpha-lipoic acid (ALA) on death and in MDA-MB-231 human breast cells.The were divided into four groups depending treatment (control, ALA, RT, ALA+RT). Cells analyzed for morphology, apoptotic mitochondrial reactive oxygen species, membrane potential, cellular senescence, cycle.Our data showed that ALA significantly promoted when combined with reflected by Annexin V staining, expression apoptosis-related factors, damages well morphological changes reduction numbers. In addition, enhanced which was shown increased HMGB1 cytosol fraction compared control, p53 activation p38 nuclear factor кB, G2/M cycle arrest.The current first report showing a new mode action (senescence induction) beyond known be RT.
