α2,3- and α2,6-linked sialic acids are important for cell binding and replication of Newcastle disease virus in chicken primary neuronal cells

ABSTRACT
Velogenic Newcastle disease virus (NDV) causes encephalitis and severe neurological disorders in avian species. Sialic acid (SA) has been considered as a receptor for NDV infection determining tissue tropism. The neurotropic mechanism of birds is completely unknown. Here we have investigated the role viral SA neurotropism chickens. We determined that α2,3- α2,6-linked receptors were implicated binding to primary neuronal cells using immunohistofluorescence virus-cell assay. Both found co-localize with velogenic strain F48E9 neuropathological lesions chicken brains after through intraocular-nasal routes. replication was more efficient than lentogenic LaSota. virus-neuronal cell capabilities both strains no difference. Furthermore, cell-binding capability significantly decreased by pretreatment sialidases cells. These results demonstrated important initiation nervous system. This study should provide preliminary evidence better understanding
