Puerarin alleviates the progression of non-small cell lung cancer by regulating the miR-342/CCND1 axis

ABSTRACT
Puerarin has recently been demonstrated to play anti-cancer roles in a series of human cancers, including non-small cell lung cancer (NSCLC), possibly through regulation cancer-related microRNAs (miRNAs).The purpose the present study was further investigate detailed role and underlying mechanism puerarin on NSCLC progression.Cell viability apoptosis were assessed using Cell Counting kit-8 (CCK-8) assay flow cytometry, respectively.Transwell assays performed determine migration invasion abilities.The qRT-PCR employed detect expression miR-342 cyclin D1 (CCND1) mRNA, CCND1 protein evaluated by western blotting.The targeted interaction between verified dual-luciferase reporter RNA immunoprecipitation (RIP) assay.We found that our data repressed viability, migration, invasion, cycle progression, enhanced cells.miR-342 overexpression hindered accelerated inhibited directly binding 3'-UTR CCND1.Moreover, overexpression-mediated anti-migration, anti-invasion, anti-cell pro-apoptotic effects abated co-transfection pcDNA-CCND1.More importantly, upregulating miR-342.Additionally, hampered progression vitro tumor growth vivo miR-342.In conclusion, suggested at least partly regulating miR-342/CCND1 axis, highlighting novel exerting property NSCLC.
