ABSTRACT
Circular RNA (circ)_0000326 has been reported in bladder cancer and cervical is concerned to be involved with the development of cancerous cells.Whereas, there have no reports concentrating on influences circ_0000326 breast (BC).Therefore, latent modulatory mechanisms BC are researched.circ_0000326 expression tissues correlative cells was evaluated via RT-qPCR, relevance between overall survival clinicopathological feature also investigated.After a series transfection, effects circ_0000326, microRNA-9-3p (miR-9-3p), Yes-associated protein 1 (YAP1) cell growth, invasion, stemness were studied by CCK-8, flow cytometry, Transwell, sphere-forming assays.The binding sites correlation miR-9-3p, YAP1 certified starBase website, luciferase reporter assay, Pearson's χ 2 test.The vivo experiment establishing subcutaneous tumorigenesis model.High-expressed discovered, which connected an undesirable prognosis.Silencing visibly inhibited MCF-7 BT549 stemness, meanwhile declining levels SRY-related high-mobility group box gene (SOX2) octamer transcription factor 4 (OCT4).miR-9-3p sponger negatively regulated circ_0000326.Moreover, confirmed as target miR-9-3p.circ_0000326 affected behaviors mediating miR-9-3p YAP1.Furthermore, silencing prohibited tumor growth vivo.The research uncovered that facilitated miR-9-3p/YAP1 axis.
